Melanotan I (Afamelanotide) is a synthetic linear analogue of alpha-melanocyte stimulating hormone (α-MSH), developed at the University of Arizona. About Melanotan I

Melanotan I (Afamelanotide) is a synthetic 13-amino acid linear analogue of alpha-melanocyte stimulating hormone (α-MSH), developed by Victor Hruby and colleagues at the University of Arizona. It was designed to be a more potent and stable version of natural α-MSH with selective MC1R agonist activity.

The key structural modifications — substitution of methionine with norvaline and replacement of L-phenylalanine with D-phenylalanine — increase receptor binding affinity, resistance to enzymatic degradation and biological half-life while maintaining the linear peptide structure.

Afamelanotide received European Medicines Agency (EMA) approval in 2014 under the brand name Scenesse (Clinuvel Pharmaceuticals) for erythropoietic protoporphyria. All Pureline Biolabs Melanotan I is independently tested by Janoshik Analytical at 99.805% purity.

Research Background

Melanotan I has been studied extensively in pigmentation biology, photoprotection and melanocortin receptor pharmacology research. This product is supplied for laboratory research purposes only.

Studies have characterised Melanotan I's binding affinity and selectivity at MC1R versus other melanocortin receptor subtypes, examining receptor activation kinetics, cAMP signalling and downstream melanogenesis cascade activation.

Laboratory and clinical studies have investigated Melanotan I's ability to increase melanin synthesis and skin pigmentation, examining the dose-response relationship between receptor activation and eumelanin versus phaeomelanin production.

Research has examined Melanotan I's photoprotective effects in UV exposure models, measuring erythema reduction, DNA damage markers and keratinocyte apoptosis — the basis for its clinical approval in EPP.

Clinical trials demonstrated Melanotan I's efficacy in erythropoietic protoporphyria, measuring pain-free time in sunlight and phototoxic reaction incidence — leading to EMA approval as Scenesse in 2014.

Reconstitution Guide

Melanotan I is supplied as a lyophilised powder and must be reconstituted with bacteriostatic water before use in research applications. Handle in reduced light conditions throughout.

Work in reduced light. Allow vial to reach room temperature. Clean the rubber stopper with an alcohol swab and allow to dry completely.

Draw required volume of BAC water into a sterile syringe. Inject slowly down the inside of the vial wall. Keep away from direct light throughout.

Gently swirl until powder fully dissolves. Solution should be clear and colourless. Store immediately at 2–8°C in reduced light.
